How can I program in a custom route to the GPS/NAV system?

The route that I have determined to be the shortest for my trip from Chesapeake Va to Shepherdstown West VA is different than the GPS's shortest route.

Is there a way to custom program in a route, my route, on the way up there?

Or perhaps a way to record the route that I take to go up there, and keep it programmed in the GPS?

Are you referring to the Nav system in the car or some other GPS device. Not sure if you're being clear enough for anyone to answer.

There's a way to have the nav take you through a series of marked points. If so inclined, you can always drop some points and have it take you through them in succession. It's been a while since I've played with that feature, but it does have it.
