The biggest thing, if you can pull it off, is to avoid stop and go traffic. My drive commute is about 35 miles round-trip. There used to always be a bad traffic jam on the way home, with several miles of stop and go traffic. Then to my amazement, CalTrans last summer actually finished a project that got rid of the jam completely. Not having to sit in that afternoon traffic jam boosted my mileage by about 25%.
Then I studied the local maps and figured out a way where I can avoid just about ALL the 'stop' points and basically take all residential streets (where it's just stop signs and surprisingly very little traffic) and cruise to work at around an average of 30-30 mph. not only do I arrive to work about 10 minutes faster than what I'd done if I sat through the major roadways' traffic jams, I actually gained a full mpg and now average mid-20 mpg! driving 30mph on residential streets! lol
